A Single Psilocybin Dose Can Reshape the Brain for a Month, Reveals Study
In a groundbreaking discovery, researchers from UC San Francisco (UCSF) and Imperial College London have found that a single high dose of psilocybin — the psychoactive compound in magic mushrooms — can induce significant physical changes in the brain that persist for up to a month.
The study, published in Nature Communications, involved 28 healthy adults who had never before used a psychedelic. Each participant was administered a 25 mg dose of psilocybin, and their brain activity and structure were monitored using EEG and MRI scans, from one hour to one month post-dose.
The researchers observed that psilocybin increased brain entropy, fortified neural connections, and enhanced participants’ sense of well-being for several weeks.
Importantly, those participants who experienced the most significant increase in flexible, varied brain activity also reported more personal insight and sustained improvements in well-being. Brain imaging one month later revealed anatomical changes, including reduced axial diffusivity in prefrontal-subcortical tracts — a structural shift associated with psychological flexibility and increased cognitive openness.
“We already knew psilocybin could be beneficial for treating mental illness. But now we have a much better understanding of how,” said lead researcher Robin Carhart-Harris. The findings suggest that the psychedelic experience itself acts as a catalyst for enduring brain health, potentially paving a new path for treating depression, anxiety, and addiction.
